实验室简介

主要研究方向:

1、 蛋白质组学新技术新方法-蛋白质高可信度鉴定、定量及翻译后修饰位点的质谱鉴定

2、 肿瘤细胞表面新生抗原和免疫多肽组的大规模鉴定及其在肿瘤发生、发展与转移中的作用机制

3、 基于肿瘤细胞表面新生抗原的特异性抗体、抗体药物偶联物、免疫多肽组的制备与体内作用机制

4、 神经系统退行性病变相关蛋白与小胶质细胞激活的调控作用机制

主要研究进展:

实验室主要利用生物质谱技术大规模鉴定与发现重大疾病发生发展相关蛋白及其动态变化,特别关注蛋白质翻译后修饰(单磷酸化及多磷酸化、乙酰化、甲基化、泛素化、糖基化、组蛋白组合修饰等)对其生物学功能的影响。针对神经系统退行性病变中小胶质细胞激活、Aβ清除与降解、神经元死亡、肿瘤细胞表面新生抗原、免疫细胞和肿瘤微环境相关巨噬细胞表面等发现并鉴定关键蛋白及其修饰变化,发现新的关键靶标和标志物;结合分子生物学、生物化学、细胞生物学和蛋白质组学技术等多重手段,利用细胞与动物模型和临床人类标本分析,对肿瘤细胞表面特异蛋白和基因融合蛋白进行体内外功能研究,同时制备特异性抗体、多肽和抗体药物偶联物,阐明药物体内调控机制。此外,我们还开展了蛋白质与抗体药物等电点、纯度、全序列、二硫键、糖基化、氧化等修饰的质谱分析研究,建立蛋白药物相关质量标准。近期主要开展了Cdk5和Sds3调控小胶质细胞激活调控神经系统炎症,以及外泌体临床检测新型标志物、结直肠癌和乳腺癌肿瘤细胞表面蛋白新靶标的大规模筛选与功能研究。
